Abstract:

Transformation of Agaricus bisporus is now well-established using Agrobacterium tumefaciens. The range of promoters used to drive transgene expression is limited at present. It is important to develop a range of promoters that can control gene expression in Agaricus bisporus during different developmental stages and nutritional conditions. This will improve the versatility of the Agaricus transformation system. We have developed an “Agaricus Molecular Toolkit” comprising interchangeable promoters and marker genes to facilitate transformation of homobasidiomycete mushrooms and subsequent analysis of gene expression. Herein, we report on the use of several promoters in both Agaricus and Coprinus when linked to several selectable and visual marker genes, along with the parameters required to successfully achieve foreign gene expression within these organisms. In particular we will report on the use of GFP as a reporter gene in Agaricus and Coprinus. Progress also will be reported on the development of genesilencing and gene-knockout technologies within homobasidiomycetes.
