Title: Genetic Variability of Flammulina velutipes and Pleurotus ostreatus Collections from Armenia

Ribosomal ITS sequences were obtained from Armenian collections of putative Flammulina velutipes and Pleurotus ostreatus in order to correctly identify the collections and to examine DNA sequence variability. Of 14 Pleurotus Armenian collections, all were P. ostreatus by sequence and were identical in sequence with the exception of three single nucleotide polymorphisms (SNPs), two of which were present in only a single collection. Of 18 Armenian Flammulina collections examined, fifteen were F. velutipes, one was an unknown biotype, one was tentatively diagnosed as F. elastica and one was diagnosed as F. rossica. Flammulina velutipes collections varied for a number of single nucleotide polymorphisms in the ITS1 region and ITS2 region and many were heterozygous for small insertions and/or deletions. For one heterozygous collection, the ITS PCR product was cloned and three haplotypes were recovered, indicating both heterozygosity of the dikaryon and heterogeneity within the ribosomal repeat. Sequence variability and heterozygosity of Armenian F. velutipes and P. ostreatus collections was higher than observed in collections from Western Europe.
