Title: Application of SE-HPLC on analyzing polysaccharides produced by mushrooms

The present work provides a way by size-exclusion HPLC (SE-HPLC) in obtaining the finger prints of exocellular soluble polysaccharides depended on their molecular size from mushrooms. The operation condition of HPLC consisted a high flow rate of deionized water at 3 ml/min through a column (1.6 I.D. x 60 cm) packed with Fructogel HW-65(S). Detection of carbohydrate was carried out by a UV-detector at 195 nm, a differential refractometer and phenol-sulfuric acid colorimetric assay for the eluent in fraction collector. Culture broth in different ages and matured fruiting bodies of Ganoderma tsugae, Lentinus edodes and Phytocordyceps ninchukispora were analyzed. The method present a fast scan on die molecular size profile and a medium size purification of polysaccharide for further bioassay and structural analysis.Please login to download the PDF for this proceeding.
