Abstract:

Putative homokaryotic cultures with isozyme patterns of type S were selected from monospore isolates by Polyacrylamide slab gel electrophoresis (PAGE) and were confirmed to produce no fruiting bodies by fruiting trials. The hybrid strain, WZU was formed by crossing between the homokaryotic cultures with different isozyme patterns of type S and 909 monospore cultures isolated from the strain were examined by PAGE esterase isozymes. It was found that the marker bands, est 1-2, est 3-4, est 30-33 were coded separately by alleles, Est Al A2, Est Bl B2, and Est Cl C2 of isozyme loci, Est A, B and C; segregation ratio of the alleles was 1:4:1, which supports the hypothesis that the random migration of pairs of meiotic products from the basidium into the spores is characteristic for Agaricus bisporus. Linkage of three Est loci was detected, R(A-B)=0.0495, R(BC)= 0.1782, R(A-C)=0.2277, the ordering of linked loci on the chromosome is Est A-B-C.
